Luke Maile was born on February 6, 1991, in Santa Clara, California. He played college baseball at the University of Kentucky, where he was a three-time All-Southeastern Conference (SEC) selection and helped the Wildcats win the 2012 SEC Tournament.

The Toronto Blue Jays selected Maile in the 24th round of the 2013 MLB draft. He made his professional debut that year and quickly rose through the Blue Jays' minor league system. Maile made his MLB debut on August 15, 2016, and played in 18 games for the Blue Jays that season.
